چکیده

Material and methods Embryos (n=120) were produced in vitro according to standardized methods and divided into three maturation groups with different insulin levels (High 10 μg/ml; Low 0.1 μg/ml and Zero=control). Gene expression data of D8BC was received through microarray studies after total-RNA-extraction at the EmbryoGENE-platform using a 2-colors-dye-swap design. Differentially expressed transcripts between control and insulin treated groups were searched by using an empirical Bayes moderated t-test in the ‘limma-package’ of R and defined as having a 1.5-fold change difference and p<0.05. Data was analyzed using IPA (www.ingenuity. com) to construct an overview over the insulin signaling pathway. Results Both genes associated with insulin resistance (PSMD4; PSMC1; MAP2K2) and its countermeasures (ADIPOR2; INSIG1) were up-regulated by insulin treatment.

P-88: Assessing Expression Changes of Some Wnt Pathway Genes During Goat Early Embryonic Development

Background: The developmental competency of embryos is affected by several factors, including the developmental pathways and their elements. In mammalian species including goat, fertilized oocyte undergoes several divisions to form a structure called blastocyst. These events depend on the successful control of temporal and spatial expression of genes involved in genome activation. One of the cr...

O-11: Diverse Effects of Polyunsaturated Fatty Acids on Oocyte Maturation and Development In vitro

Background: Polyunsaturated fatty acids (PUFAs) have been shown to influence fertility and endocrinology of reproduction and metabolic activity in many species. In dairy cows, we and others have shown changes in steroid and metabolic hormones and prostaglandins leading to alteration of ovarian activity and uterine function. These can influence fertility by changes in folliculogenesis cyclicity ...

Insulin exposure during in vitro bovine oocyte maturation changes blastocyst gene expression and developmental potential.

Metabolic imbalance impairs fertility, because changes in concentrations of metabolites and hormones in the blood and follicular fluid create an unfavourable environment for early embryonic development. Insulin is a key metabolic hormone known for its effects on fertility: insulin concentrations are increased during energy balance disturbances in diabetes or metabolic syndrome. Still, insulin i...

The Effects of Progesterone on Oocyte Maturation and Embryo Development

Oocyte maturation and embryo development are controlled by intra-ovarian factors such as steroid hormones. Progesterone (P4) exists in the follicular fluid that contributes to normal mammalian ovarian function and has several critical functions during embryo development and implantation, including endometrial receptivity, embryonic survival during gestation and transformation of the endometrial...
